Learning outcomes

Aim of the course: give an overview of the principles and purposed behind the study of functional proteomics and to debate over the main methodological strategies in the field, together with up to date literature on the subject.

Syllabus

The general concept of functional proteomics.
Main proteomic workflows: top down vs bottom up.
Insights into protein-protein functional interactions.
Physical methods: Calorimetry
Surface immobilized methods: QCM, Protein arrays, iSPR
Prey strategies: label vs label free.
Bait strategies: proteins, motifs, epitopes, peptides, combinatorial libraries, synthetic receptors.
Affinity purifications, tandem affinity purifications, coimmunoprecipitation, pulldown, crosslinking.
Analytical ultracentrifugation.
Approaches to study SS bonds
Structural information on the protein interactions
post translational modifications (phosphorylation)
Peptidomic
In vivo validation: two hydrib, FRET, BRET
