Learning outcomes

Fornire allo studente le basi per comprendere le possibilità di sfruttamento biotecnologico di organismi microbici quali produttori di nanomateriali con particolare riferimento a nanoparticelle metalliche

Syllabus

The course is aimed to give knowledge and abilities about the use of microorganisms, either bacteria and fungi, for the biosynthesis of nano-structures, especially metal nano-composites. Thus the course has the object to give the basic information to understand the biosynthetic mehanisms of metal nano-cpmposites in microbial cells and to analyze the protocols for the synthesis of this nano-materials by microorganisms. Moreover knowledge will be give about main biomolecular techniques to apply for microbiology.

Lectures
The microbial cell - The microbial diversity - Structures and funcions of the bacterial cell - Microbial Growth - Resistance Mechanisms to metals and metalloids in bacteria - Production of metal nanoparticles/nanorods by bacteria and fungi: Mechanisms of synthesis and potential applications. We will consider: i) selenium; ii) tellurium; iii) silver; iv) gold; v) magnetic nanoparticles and magnetotactic bacteria.
Laboratory
First Part
- Using bacterial cells for the synthesis of selenium and tellurium nanoparticles
- extraction and purification of metal biogenic nanoparticles from microbial biomass
- Analysis of aantimicrobial and antibiofilm activities of the biosynthesized nanoparticles
Second Part
- Molecular Biotechniques: i) DNA aextraction from bacterial cultures; ii) DNA amplification through PCR; iii)electrophoresis on agarose gels; iv) cloning

Assessment methods and criteria

The testing of the learning will be carried out through an oral exam. The student has to prepare a case study on the basis of one or more scientific papers discussed during the lectures. The case of study will be the starting point to verify the comprehension of the mechanisms of biogenic synthesis of metal nanoparticles in microbial cells and the main applications of the biosynthesized nanoparticles, with particulr reference to the metals and metalloids considered during the lectures. Moreover the comprehension of molecular techniques applied during practical class will be verify as well.
